I find it funny that scientists study the brain and neurotransmitters and can explain the basics but have absolutely? no clue what goes on in there in more detail and then make medication that can help (for ex. with depression) but do not do so on more microscopic level

I think it is illustrating conduction down the neurone, and how? there is a net movement of positive charge into the axon as the action potential is propogated. This is because sodium influx (sodium ions carry a single positive charge) is greater than potassium efflux ( potassium ions are positive also). It is also important to note that it is illustrating an unmyelinated neurone, as it occurs across the entire length of the axon. Hope that helps :) x

It depends on the ion channel it opens. If it opens a Na+ channels, that causes an excitatory signal. If it opens a Cl- channel, that causes an inhibitory signal. GABA is usually an inhibitory Neurotransmitter. Dopamine can be either excitatory? or inhibitory. However, they have different receptors that can open different channels that allow different ions to flow in. This can result in an excitatory or inhibitory signal. Hope that helps.

Hello, I liked your video, it explains things simply and clearly.I? have a question though: When a signal traveling on the axon reaches the synapse a neurotransmitter is released.This can be let's say dopamine or GABA or smth else, and is either inhibitory or excitatory, correct?Then the receiving neuron binds this neurotransmitter with its receptors and sends an electrical signal to the core of the cell, am I correct?The question is what kind of signal is this one - excitatory or inhibitory?10x

I don't have OCD but, more like perfectionism. Oddly, I know Im not perfect and don't try to be. What has to be perfect is the things around me. When I have something thats supposed to be symetrical, it HAS to be symetrical. If it's not, I try to fix it. If I can't fix it, I get rid of it. It's the same with grammar and spelling. I had a weird OCD too. I was afraid people stole intelligence from me, that they absorbed my intellect and used it to their advantage. So what I would do was to ask myself questions of subjects I reviewed like 8 years ago, and if I didn't remember them very well I would look em up in a book or on the internet when I? got home to make sure I kept my intellect. It was really frustrating, I always thought I was getting dumber and people around me were getting smarter out of my own capacity.

very cool. I'm old. Like - really old - 68. (yeah - you'll get there too someday. you never figure it'll happen, but it does) I struggled with this stuff when I was a kid, too, before anybody ever knew ANYthing about it. I just thought I was crazy. It scared the shit out of me. I've spent my whole life, still do, studying the brain so I could learn how it all works & live better/ not hurt myself or anyone else. Been? I also get irate at those people who use the term "OCD" lightly. I used to be really bad in high school. I was always afraid of getting other people sick. I didn't touch hands, hug, or even borrow pencils. I picked a pencil up from the floor one day and I could not think until I went to wash my hands. Anything that binds to a receptor and activates it is classed as an 'agonist'. So something that would be an antagonist for this would be something that binds to this receptor? and does not activate it, thus blocking the agonist. An antagonist in this case would stop GABA binding and therefore stop chloride ions passing across the membrane, preventing hyperpolarisation. To your second point the action potential is not continuing, the 'post-synaptic' effect is hyperpolarisation.

I agree with you in every way; so many drugs are overprecribed. Various SSRIs are prescribed for depression of any kind when they were showed to be clinically insignificant except in severe depression.And they don't stress the risks of long term benzo use. If I find any conclusive info. on the neurochemistry of benzodiazepine dependence, I will let you know.

I (and many others) would be profoundly grateful for any knowledge you could help with. This is a huge sticking point, as most GP's & psychiatrists haven't a clue as to why benzodiazepine withdrawal syndrome, or protracted withdrawal syndrome occurs after a rapid taper from benzos, which more often than not,? lasts for years. Unfortunately, over 100 million prescriptions are written yearly in the US (a DEA stat), which is approaching insanity in my opinion. But sincere thanks!

so you may be partially correct in that gene expression could be changed, causing seriously low GABA production, although I doubt that is the case as synthesis of neurotransmitters is relatively "simple" and very controlled. All of this is speculation and I will need look at some literature. Antibodies with? an attached fluorophore can be used to seek out specific enzymes. If a brain of an benzodiazepine dependent person was stained with a certain antibody, one could look at GAD levels.

Epigenetics displays great promise in explaining this phenomenon, as changes in gene-expression are often the long-term regulators of many neurochemicals. I would guess that maybe changing GAD gene expression (GAD is the enzyme that synthesizes GABA from glutamate) or expression of some? other GABAergic constituent might cause long-term dysregulation .

Benzodiazepine dependence is caused neurons decreasing their concentration of GABA receptors (decreased inhibition), preventing chloride ion flow; however, NMDA receptors are upregulated to a lesser extent (increased excitation) lending to the drug's withdrawal syndrome. This glutamate rebound can also cause excitotoxicity in theory, and? excitotoxicity does happen during alcohol withdrawal because alcohol primarily antagonizes NMDA while agonizing GABA to a lesser extent.

To be? clear, I've personally experienced detox from years of pre/post surgery morphine & other opioids (one to two week recoveries), but the return to a psychological baseline after years of benzodiazepine use in half of we long-time users has taken many months or years. the post-synaptic effect is inhibition- meaning that the likelihood of the post-synaptic? cell producing an action potential is reduced (Chloride enters the cell, altering the resting potential of the cell). Remember- agonists are ligands that bind and produce an effect, whereas antagonists are ligands that bind and produce no effect. Hope that helps.
